The Foodborne Toxin Detection and Prevention Research Unit located in Albany, CA is seeking a Research Molecular Biologist (Research Associate). This is a full time, temporary appointment Not-to-Exceed 2 years. The mission of this Unit is to conduct research to enhance food safety and biosecurity by developing new methods for analysis of toxins and bacteria in foods, and new strategies to reduce or eliminate mycotoxin contamination of agricultural commodities (especially tree nuts and figs). We validate these technologies and transfer knowledge to the scientific community, the food industry, and regulatory agencies responsible for the safety of the U.S. food supply.

The incumbent will clone and sequence selected mouse anti-Shiga toxin monoclonal antibodies and graft selected CDRs onto the most similar human IgG germline framework. Phage display mutant libraries will be created using the top CDR-grafted murine-human chimeric scFv as the parental antibodies through Kunkel-like mutagenesis approach for affinity maturation. Mutants with highest affinity will be cloned into appropriate expression vectors containing human IgG constant regions for production of full size human IgGs in mammalian cells. The binding affinity, prophylactic and therapeutic efficacy of the humanized antibodies will be evaluated in vitro and in vivo.
